TUMORI. CONFERMATA ORIGINE VIRALE DI MOLTI TIPI DI CANCRO: STUDIO TEDESCO HA TROVATO TRACCE DI VIRUS NEL 13% DEI 2600 CASI ESAMINATI
11 febbraio – Si sapeva già che alcune infezioni virali sono in grado di favorire lo sviluppo di tumori. Il virus dell’epatite B, il virus di Epstein- Barr e i papillomavirus ad esempio, sono correlati rispettivamente ai tumori del fegato, a tumori allo stomaco e linfomi e ai tumori di testa, collo e cervice uterina. Ora un nuovo studio condotto dai ricercatori del German Cancer Research Center ha analizzato la presenza di virus in 2600 campioni di tumore alla ricerca di nuovi virus potenzialmente oncogeni e ha scoperto tracce di virus nel 13%dei casi.
Quali virus sono associati all’insorgenza del cancro? In che modo le infezioni possono favorire lo sviluppo di tumori? Se si riuscisse a rispondere con precisione a queste domande, si potrebbero prevenire alcuni tumori semplicemente con un vaccino.
Ad oggi sappiamo che diversi virus, in concomitanza o meno con fattori ambientali, sono la causa di tumori: il virus di Epstein-Barr (EBV) aumenta il rischio di sviluppare alcuni tipi di cancro come i linfomi; il virus dell’patite B (HBV) e C sono associati al 70/ 85% dei casi di epatocarcinoma; i papillomavirus sono invece associati al cancro della cervice uterina, c’è poi i virus della leucemia a cellule T di tipo 1.
Per identificare altri tipi di virus gli scienziati del German Cancer Research Center hanno studiato sistematicamente il Dna di oltre 2600 campioni di tumore da pazienti affetti da 38 tipi diversi di cancro. Hanno osservato tracce di virus nel 13% dei casi e sono riusciti ad identificare alcuni dei meccanismi messe in atto dai patogeni che portano a mutazioni cancerogene del Dna dell’ospite. Il rapporto dei ricercatori è stato pubblicato sulla rivista Nature Genetics, il lavoro fa parte dell’analisi Pan-Cancer of Whole Genomes (PCAWG), un’iniziativa lanciata dall’International Cancer Genome Consortium (ICGC).
I ricercatori hanno effettuato un’analisi bioinformatica completa dei dati provenienti dal sequenziamento dei genomi tumorali. Hanno scoperto le tracce di 23 tipi di virus in totale, in 356 pazienti. Come previsto, hanno identificato il virus di Epstein-Barr, responsabile in particolare di linfomi e carcinomi gastrici e rinofaringei: era presente nel 5,5% dei genomi studiati. Il Dna dell’epatite B, invece, è stato trovato in 32 dei 330 casi di cancro al fegato e il papillomavirus in 19 dei 20 casi di carcinomi cervicali studiati e in 18 dei 59 casi di tumori della testa e del collo. Il team ha individuato altri virus noti per la loro associazione con lo sviluppo di tumori, come un retrovirus nel carcinoma renale. Lo studio non ha portato alla scoperta di virus completamente sconosciuti.
Il modo in cui i virus riescono a trasformare le cellule e ad esercitare un potere oncogeno variano da un ceppo all’altro. Il meccanismo più comune osservato dai ricercatori è l’integrazione del genoma del virus all’interno del Dna della cellula ospite. Se il virus si inserisce nel punto giusto, questo provoca delle mutazioni cancerogene, come succede con i papillomavirus e con il virus dell’epatite B. “Abbiamo spesso osservato l’integrazione del Dna dell’HPV nel promotore della telomerasi: questo interruttore genetico guida la produzione della telomerasi, conosciuta come l’enzima dell’immortalità, mutato in molti tipi di cancro”, ha spiegato Marc Zapatka, autore principale dello studio.
A volte invece, è la risposta delle cellule che si difendono dall’infezione virali a causare le mutazioni genetiche alla base dei tumori. Per attaccare il virus infatti, la cellula produce degli enzimi, detti APOBEC, che però, se non sono regolati correttamente, possono provocare mutazioni del Dna della cellula stessa.
L’organizzazione Mondiale della Sanità (OMS) stima che oltre il 15% di tutti i tumori sia direttamente o indirettamente attribuibile a patogeni infettivi. L’Agenzia internazionale per la ricerca sul cancro (IARC) di Lione ha classificato 11 agenti patogeni diversi – virus, batteri e vermi – come cancerogeni e stima che un tumore su 10 sia collegato a virus. In questo studio i ricercatori hanno scoperto “tracce di virus in un numero considerevolmente maggiore di tumore, rispetto a quanto osservato negli studi precedenti, che si erano basati esclusivamente sull’analisi dell’RNA”, ha osservato Peter Lichter, autore senior dello studio. “Cionostante – ha detto – non siamo stati in grado di confermare la speculazione comune secondo cui altri virus, ancora sconosciuti, siano associati al cancro.”
